The first demonstration of laser action in ruby was made in 1960 by T. H. Maiman of Hughes Research Laboratories, USA. Many laboratories worldwide began the search for lasers using different materials, operating at different wavelengths. In the UK, academia, industry and the central laboratories took up the challenge from the earliest days to develop these systems for a broad range of applications. This historical review looks at the contribution the UK has made to the advancement of the technology, the development of systems and components and their exploitation over the last 60 years.
Strategies for rapid rebreeding of lactating ewes in the spring

Rapid rebreeding of winter- and spring-lambing ewes is essential if ewes are to lamb more than once per year, but fertility of lactating ewes is often low and early weaning of lambs may be undesirable in forage-based production systems. Selection to improve fertility in spring matings has been successful and led to development of ewes with a reduced seasonal anestrus. Potential for rapid rebreeding of lactating out-of-season breeding (OOS) ewes was tested in three studies. In Experiment 1, effects of short-term lamb removal on rebreeding was evaluated over 2 years using 71 January-lambing OOS ewes. At an average of 63 days postpartum, 36 ewes had lambs removed for 72 h, and all ewes were joined with rams. Circulating progesterone levels indicated that 74% of ewes ovulated before lamb separation; 91% of ewes mated within 5 weeks of ram exposure, 85% were diagnosed as pregnant and 75% lambed. The average interval between lambings was 225 days. In contrast to results observed in cattle, none of the measured variables was affected by lamb separation (P>0.20). Experiment 2 compared rebreeding performance of 24 OOS and 23 St. Croix ewes that lambed in January and averaged 60 days postpartum at ram introduction. More OOS ewes ovulated, mated and became pregnant during the first 21 days of ram exposure (83.3%, 58.3%, and 41.7%, respectively; P<0.001) compared with St. Croix ewes (26.1%, 0%, and 0%, respectively). After 39 days of ram exposure, pregnancy rates still favored OOS ewes (66.7% v. 39.1%; P=0.06), but the percentage of ewes that lambed did not differ (P>0.20) between OOS (47.8%) and St. Croix ewes (34.8%). In the third study, 34 March-lambing OOS ewes were exposed to rams on May 3 at an average of 40 days postpartum to characterize their reproductive performance. After 39 days of ram exposure, 52.9±8.7% of the ewes had mated, and 38.2±8.5% were diagnosed as pregnant. However, only 20.6±7.0% of the ewes produced viable lambs, suggesting a high level of uterine insufficiency. Spring fertility of lactating OOS ewes in these studies was one of the highest reported in the literature and indicated that selection for fertility in spring mating would improve reproductive performance in accelerated lambing programs. However, exposure of lactating OOS ewes to rams at 30 to 50 days postpartum was associated with high prenatal lamb mortality.

Data from black fly traps in Deep River over the past 5 years indicate that larvicides applied to breeding sites in streams surrounding the town have substantially reduced the numbers of this pest. Very high numbers occurred in 1972, the result of invasion associated with high westerly and northerly winds at a critical period of time or with heavy protective snow cover during the preceding winter. The lowest population levels occurred in 1975, when treatment included a stream near the town which had not been treated previously. The results indicate that with proper stream management it is possible to substantially reduce the nuisance value of black flies in a small northern town.

Blood-meals of two species of haematophagous insects, Glossina morsitans Westw. and Aedes (Stegomyia) aegypti (L.), have been ‘ marked ’ by allowing groups of adults to feed on an ox to which an aqueous solution containing 4 g. of trypan blue had previously been administered intravenously. The dyestuff is detected by a paper chromatographic procedure in which the blood-meal, mixed with 0·1N NaOH, is applied to a narrow strip of Whatman No. 1 chromatographic filter paper and the chromatogram is developed in 0·1N NaOH. Under these conditions, trypan blue remains at the origin, whereas the other coloured components of the blood-meal move away from the point of application. The duration of the mark in the ox and in the insects has been examined. The trypan blue was detected in four out of six fresh blood-meals of Glossina taken 38 days after injection of the dye into the ox, in six out of six examples of Glossina examined two days after blood-meals taken 24 days after the injection, in four out of six examples of Glossina examined eight days after blood-meals taken immediately following the injection, in six out of six examples of Aedes one day after blood-meals taken 14 days after the injection and in six out of six examples of Aedes two days after blood-meals taken immediately following the injection.

In a factory population the occurrence of reactions to tetanus toxoid was recorded after 6740 injections. The incidence of general reactions was 0·3 % and of local reactions 2·6%. The local reaction rate to the first injection of the basic immunization course was 0·9%, to the second injection 2·7%, and to the third injection 7·4%. To booster injections the rate was 1·6%. The local reaction rate was appreciably higher in women than in men – 14·4 % and 5·7 % respectively in the case of the third injection – and the incidence among women increased with age.
Tetanus vaccine containing 10 Lf of toxoid caused fewer reactions than one containing 20 Lf, but a reduction in the content of aluminium adjuvant did not affect the reaction rate.
Almost all reactors were found to have a satisfactory serum antitoxin concentration at the time of the reaction or developed a satisfactory immunity within 1–6 months.
Skin tests were made in 32 hypersensitive patients. Neither the diluent, thiomersal preservative, nor the culture medium appeared to be responsible for hypersensitivity. The degree of hypersensitivity elicited by a special highly purified toxoid was only very slightly less than that elicited by the commercially pure toxoid. It is suggested that reactions are largely due to the toxoid antigen itself rather than to impurities or other components of the vaccine.

Of the surface antigens identified by radio-iodination, two-dimensional gel analyses showed no similarities between those of Schistosoma haematobium and Schistosoma mansoni, thus providing a basis for the species specificity of these antigens described previously (Simpson, Knight, Hagan, Hodgson, Wilkins & Smithers (1985) Parasitology 90, 499–508). The surface antigens of S. haematobium were glycosylated and comprised an acidic polypeptide of Mr 17000 as well as a complex set of polypeptides of approximate pI 6–7, which resolved in the Mr range 20000–30000. At least one of the lower Mr forms of this complex is also present in the adult worm. Limited cross-reaction was observed with S. mansoni infection sera and this may be due to a shared carbohydrate epitope. In contrast, extensive cross-reaction was observed using sera from mice immunized with S. bovis. This pattern parallels the species-specificity of vaccine-induced immunity. Extensive cross-reaction was also observed within cell-free translation products of m-RNA from adult worms of S. haematobium and S. mansoni by use of heterologous human infection sera. The few antigens which were species-specific may represent surface antigens.

Surface antigens of Schistosoma haematobium were identified by 125I-surface labelling of schistosomula followed by immunoprecipitation of the solubilized, labelled surfaces. The major antigens, after electrophoresis, formed a continuous smear corresponding to a molecular weight in the range 35–24000; in addition, a 17000 antigen was also identified. These surface antigens, in contrast to somatic antigens, were species-specific, as judged by immunoprecipitation with human anti-S. mansoni serum and serum from mice vaccinated with highly irradiated S. mansoni cercariae. S. haematobium surface antigens, however, were recognized to some extent by serum from mice chronically infected with S. mansoni. It is suggested that this cross-reactivity may reflect the heterologous immunity demonstrated experimentally between these two species, whilst the species-specificity of vaccine sera to surface antigens may mirror the highly specific immunity induced by vaccination.

In this study, 82 sex offenders incarcerated at the Massachusetts Treatment Center were administered a brief structured clinical interview focusing on caregiver and institutional history up to age 18. Interview data were examined along with archival information coded from the clinical files. Four factors were derived from principal components analysis and were entered into a series of multiple regression analyses. There were two noteworthy results. First, sexual and nonsexual aggression in adulthood each were related to distinct aspects of developmental history. Whereas caregiver inconstancy and sexual deviation in the family wére related to the severity of sexual aggression, childhood and juvenile institutional history and physical abuse and neglect were associated with severity of nonsexual aggression. Second, contrary to previous studies of other criminal populations, the severity of aggression rather than frequency of crimes was predicted by developmental history. The results of the study suggest that childhood experiences with and disruption of caregiver relationships may be important to understanding sexual aggression in adulthood.
Lactation failure in crossbred Sahiwal Friesian cattle

Milk producers in Malaysia make extensive use of crossbred Sahiwal Friesian dairy cattle. These animals have, however, been found susceptible to lactation failure. A survey of cows in an experimental herd of F1 Sahiwal Friesian animals indicated that, in 30% of animals, milk yield decreased to negligible levels within the first 8 weeks post partum. Lactation failure was associated with a progressive increase in the amount of residual milk left in the udder after normal milking. By week 3 of lactation, residual milk volume was significantly greater than that in animals that, based on previous lactation history, were not susceptible to lactation failure, and accounted for up to 30% of milk available at the morning milking. The cellular consequences of residual milk accumulation were evident in the activities of acetyl-CoA carboxylase, fatty acid synthetase and galactosyltransferase, key enzyme markers of cellular differentiation, which decreased in glands undergoing lactation failure and were lower than values measured in tissue of control cows. Mammary cell number, estimated by tissue DNA content, was also reduced in animals undergoing lactation failure. These indices of mammary development indicate that lactation failure is the result of premature involution in susceptible animals. Premature involution is a predictable consequence of progressive milk stasis in failing lactation, and attributable to an increase in autocrine feedback by inhibitory milk constituents. The progressive increase in residual milk is, on the other hand, unlikely to be attributable to impaired mammary development. Measurements of milk storage during milk accumulation showed no differences between control and lactation failure cows in the distribution of milk between alveolar and cisternal storage compartments. We conclude that lactation failure in Sahiwal Friesian cows is due to a failure of milk removal, and probably the result of an impaired milk ejection reflex rather than to the glands' milk storage characteristics.
Partitioning of milk accumulation between cisternal and alveolar compartments of the bovine udder: relationship to production loss during once daily milking

Experiments were undertaken to validate a method (using adrenaline injection) for determination of the size of cisternal and alveolar compartments in the udder, to use this method to determine the pattern of milk accumulation in the udder over time and to determine the relationship between the size of the alveolar and cisternal compartments and tolerance of once daily milking. Cows received intrajugular injections of adrenaline (3 mg) immediately before milking, to block milk ejection and allow harvesting of the cisternal milk fraction. This was followed by removal of the alveolar fraction 30 min later after intrajugular oxytocin (5 i.u.) injection. Results obtained were similar to those obtained by catheter drainage. The alveolar compartment was 90% full at 16 h post milking while the cisternal compartment filled more slowly and was only 70% full at 24 h post milking. At full capacity (measured at 40 h), the volumes of milk contained in the cisternal and alveolar compartments were similar. In a further experiment involving identical twin cows, it was shown that the greater the degree of filling of the cisternal compartment at 24 h, the lower was the production loss on once daily milking. This suggests that the freedom of the alveoli to drain was an important factor in the production loss on once daily milking. Although there were significant correlations within twin sets for milk yield and the size of udder compartments, the relationship within twin sets for yield loss on once daily milking was not statistically significant.
Defect Tails in GE Implanted Si Probed by Slow Positrons and Ion Channeling

Positron annihilation spectroscopy has been used to profile the distribution of defects following implantation of 120keV Ge+ into (100) Si in the dose range l x 1010 - lx104 cm−2 . The openvolume defect profiles can be adequately fitted assuming a simple rectangular block distribution extending to 350nm. Using anodic oxidation and etching, a procedure is described which allows details of the defect tails beyond the range of the implanted ion, usually inaccessible to positron -2 annihilation measurements, to be determined. For a time averaged dose-rate (Jr) of 0.02μA cm−2 and incident angle of 7°, open-volume defects are found to exist at concentrations exceeding 1016cm−3 at depths upto 600nm whereas the peak of the depth distribution of the implanted ions (Rp) is 76nm, measured using SIMS. When the time-average dose-rate is increased by a factor of 10, defects persist at concentrations in excess of 1017cm−3 beyond lμm and the Rp increases to 101nm. The open-volume defect profiles are compared to those deduced from Rutherford backscattering-channeling using the fitting routine DICADA.
Changes in casein composition of goats' milk during the course of lactation: physiological inferences and technological implications

Five British Saanen goats were milk sampled during the first 39 weeks of lactation to determine changes in casein composition. Caseins were separated by anion- and cation-exchange FPLC to determine the relative amounts of the individual caseins. Acid, alkaline and SDS-PAGE were used to determine possible genetic polymorphisms and observe any lactational changes. Total casein nitrogen was determined using a micro-Kjeldahl method and this allowed the concentrations of individual caseins to be calculated. The milk of one animal, which had the deduced genotype αs1-CnAB, showed higher concentrations of both total and αs1-casein. The remainder of the group were either heterozygous αs1-CnBE or, more probably, homozygous αs1-CnE and produced milk of a generally lower protein concentration. Both FPLC and PAGE results showed that the relative amounts and concentrations of αs2-casein decreased with stage of lactation, consistent with its susceptibility to proteolysis. The relative amounts of the breakdown products of plasmin attack on β-casein, γ-caseins, were highly negatively correlated with milk yield (r = –0·942, P < 0·001) in the declining phase of lactation, reflecting the gradual involution of the gland at this time. The relative amount of κ-casein increased by ∼ 50% after peak lactation and its concentration almost doubled near the end of lactation. These compositional changes may alter the processing qualities of goats' milk in relation to cheese production.

Milk yield is ultimately constrained by the number of mammary secretory cells and the amount produced by each cell; increasing yield during early lactation is associated with increased output per cell, whilst decreasing yield during declining lactation correlates with reduced number of cells. Overall, yield is highly correlated with mass of secretory tissue but strategic control of lactation involves a galactopoietic complex of hormones including GH, prolactin and oxytocin acting to ensure efficient milk ejection and maintenance of secretion. Fine control, on the other hand, is achieved locally within the mammary gland by an autocrine mechanism which matches supply of milk to demand. The feedback inhibitor of lactation (FIL) is a milk protein which is inhibitory to secretion, so as milk accumulates between milkings, secretion rate gradually falls. The more frequently FIL is removed by milking, the greater the overall secretion rate. Storage of milk occurs within secretory tissue (alveolar milk) but also in the cistern (cisternal milk); FIL is effective in alveolar milk but not in cisternal milk, because it is then remote from its site of action. Therefore predictions would be that for a given mass of secretory tissue, large-cisterned cows should produce more milk, be more tolerant of infrequent milking but be less responsive to frequent milking. Methods developed by us for determining cisternal and alveolar milk storage spaces have obtained statistically proven support for the latter two predictions in dairy cows; the first prediction has been proven in goats. It has been shown that cisternal milk fraction increases during the course of lactation and with increasing parity. It is now intended to investigate the feasibility of incorporating storage characteristics into future selection strategies.
Relationship between milk storage characteristics and the short-term response of dairy cows to thrice-daily milking

Twenty lactating dairy cows were used to investigate the relationship between the site of milk storage in the udder and the short-term response to thrice-daily milking. Cisternal and alveolar milk volumes were measured 8 h after an ordinary morning milking by catheter drainage and machine milking with oxytocin respectively. The response to thrice-daily milking was assessed using a half-udder technique and the relative milk yields quotient (RMYQ). Over the first 7 days, both halves were milked twice daily (8/16 h intervals) and milk yields over the final 4 days of this period were higher for left fore/right hind (LF/RH) (12·4 (s.e. 0·85) kg/day) than for RF/LH (10·5 (s.e. 0·63) kg/day) which was milked after LF/RH throughout the experiment. Over the following week, LF/RH quarters were milked an additional time (8/8/8 h intervals) and yields over the final 4 days were increased (15•7 (s.e. 0·95) kg/day) compared with control quarters (9·8 (s.e. 0·73) kg/day). In a final 4-day period, animals were milked twice daily and half udder yields were 13·1 (s.e. 0·89) kg/day and 10•6 (s.e. 0·77) kg/day respectively. Differences between yields from the two halves of the udders were highly significant in all 3 weeks of the experiment (P < 0·001). Cistern milk yield as a proportion of total milk yield at 8 h (cistern proportion) averaged 0·170 (s.e. = 0·0275; range 0·020 to 0·334) and tended to be greater for multiparous (0·215, s.e. 0·0279) than for primiparous animals (0·118, s.e. 0·0437; P = 0·076). During the periods of twice-daily milking, the proportion of milk yielded from LF/RH quarters was not significantly related to cistern proportion (P = 0·70 and 0·43 for weeks 1 and 3 respectively). However the response to thrice-daily milking, assessed as RMYQ, was significantly related to cistern proportion both when changing up to, and down from, thrice-daily milking (P < 0·01). Animals with low cistern proportions showed larger responses to thrice-daily milking. There was a significant relationship (P < 0·05) between the responses on changing up to, and down from, thrice-daily milking. Primiparous animals tended to exhibit smaller declines on returning to twice-daily milking than multiparous animals with equivalent responses to thrice-daily milking.

The objective of this work was to investigate phenotypic sources of variation in the sites of storage of milk within the udder. Ten primiparous and 10 multiparous dairy cows were studied over two successive lactations in order to investigate the effects of age, parity and stage of lactation. Cistern and alveolar milk volumes were estimated at a standard 8 h after a normal morning milking by catheter drainage and machine milking with oxytocin respectively. Measurements were made at approximately 9, 20, 30 and 42 weeks of the initial lactation and weeks 1, 4, 7, 10, 20 and 34 of the subsequent lactation. In the initial lactation, there was a significant difference between primiparous and multiparous animals in the rate of decline of total daily milk yield (P < 0·001). Cistern milk yield (kg/8 h) from multiparous animals declined over the lactation whilst that for primiparous animals was maintained. The effect was shown as a significant interaction effect (P < 0·05). For both multiparous and primiparous animals cistern proportion (cistern yield divided by total milk yield at 8h after a normal milking), increased over the lactation (P < 0·001). This was largely a consequence of the reduced total milk yield. Eleven animals were studied through the subsequent lactation. Cistern yield followed a lactation curve of similar form to that for total yield. The decline in cistern yield over this lactation was much greater than the decline with multiparous animals in the previous year. This may, in part, reflect a faster decline in total daily yield. Stage of lactation and parity must be taken into account if a standardized evaluation of cistern characteristics is to be obtained.
Metabolic adaptations in goat mammary tissue during long-term incomplete milking

Lactating goats were milked incompletely in one gland and normally in the other gland twice daily for 24 weeks. Yields of the incompletely milked glands were adjusted so that a calculated 100 ml milk remained in the gland at the end of milking. On average this residual volume was greater by 7·6% of the total milk volume than that left after normal milking and was 25·9% greater than the capacity of the alveoli. After 24 weeks, the weekly milk yield of the incompletely milked gland was 23·8% lower than that of the other gland (P < 0·001) when expressed in relation to cell number. The metabolic capacity of secretory cells in the two glands was measured in serial tissue biopsies. Two weeks of incomplete milking had no effect on enzyme activities or metabolic fluxes in the incompletely milked gland, but after 24 weeks the activities of several key enzymes were significantly lower in this gland. This was associated with a reduced rate of synthesis of total protein and casein in expiants freshly prepared from the tissue. The rate of lactose synthesis also tended to be lower in expiants from the incompletely milked gland. Secretory cell number in the two glands was similar at the end of incomplete milking. The data indicate that incomplete milking over a long period caused partial secretory cellular involution via a local chemical feedback mechanism in the gland.
Digestion and synthesis in the rumen of sheep given diets supplemented with free and protected oils

1. Six wether sheep were each provided with a permanent cannula in the rumen and re-entrant cannulas in the proximal duodenum.
2. In a preliminary study, the sheep consumed 200 g hay and 400 g concentrates supplemented with up to 40 g linseed oil, coconut oil or cod-liver oil daily. Feed was refused at higher levels of supplementation.
3. Five of the sheep were used in a 5 × 5 Latin-square experiment. They were given 200 g hay and 400 g concentrates alone (B) or supplemented with 40 g linseed oil (L), coconut oil (C), protected linseed oil or protected coconut oil daily. The protected oils were prepared by emulsifying the free oils with formaldehyde-treated sodium caseinate. Formaldehyde-treated sodium caseinate was also included in the other three diets.
4. Digestion in the stomach was measured by spot sampling duodenal digesta, using chromic oxide-impregnated paper as the marker. Microbial flow at the duodenum was measured by use of both diaminopimelic acid (DAPA) and RNA as microbial markers.
5. Both the free oils had broadly similar effects despite their very different fatty acid compositions. Digestion in the stomach of organic matter (OM) was reduced from 0·48 (diet B) to 0·29 (diets L and C) and that of neutral-detergent fibre from 0·50 (diet B) to 0·19 (diet L) and 0·12 (diet C). The molar proportions of acetic acid and n-butyric acid were decreased and that of propionic acid was increased. Protozoal numbers were reduced by 78% (diet L) and 90% (diet C). The flow of total nitrogen and microbial N was increased by both oils and the efficiency of microbial protein synthesis (g N/kg OM apparently digested in the rumen) was increased from 30 (diet B) to 85 (diet L) and 74 (diet C) when based on DAPA and from 41 (diet B) to 94 (diet L) and 81 (diet C) when based on RNA. The efficiency when based on true digestion of OM (g N/kg OM truly digested in the rumen) was increased from 23 (diet B) to 46 (diet L) and 44 (diet C) when based on DAPA and from 29 (diet B) to 49 (diet L) and 46 (diet C) when based on RNA. The amounts of microbial OM (g/d) at the duodenum were increased from 68 (diet B) to 124 (diet L) and 106 (diet C) when based on DAPA and from 92 (diet B) to 136 (diet L) and 115 (diet C, non-significant) when based on RNA.
6. When the oils were given in the protected form, the effects on digestion in the stomach were reduced but not eliminated. No significant increases in the amount of total N or microbial N at the duodenum were established, though there was a tendency for an increase in the efficiency of microbial protein synthesis with protected linseed oil. The results suggested that the method of protection used reduced the effects of the oils on rumen digestion and synthesis but was only partially successful in preventing hydrogenation of the fatty acids.
7. It is concluded that free oils can markedly increase the efficiency of microbial protein synthesis, possibly by their defaunating effect, and that this may enhance the potential for using non-protein-N on oil-supplemented diets.

A randomized controlled study of 38 catchment unit psychiatric in-patients in a discussion group showed no significant additional benefit compared with 36 in-patients in an activity group. Symptom change scored by three rating groups agreed closely, average r = +0.64. Patients treated with ECT showed 2.6 times as much reduction in morbidity as those not so treated, but there was still no appreciable difference between discussion and activity groups when this was allowed for. Other treatment factors and the Hawthorne effect appeared to obscure any effect due to group therapy.
